PT-141 calculator
PT-141 is bremelanotide, the drug sold as Vyleesi. The FDA dose is 1.75 mg subcutaneous at least 45 minutes before sex, no more than one dose in 24 hours and no more than 8 a month.
- Frequency
- as needed, no more than once in 24 hours; label caps at 8 doses a month
- Timing
- 45 minutes to 4 hours before; effects last several hours and can carry into the next day
- Cycle
- as needed only; no daily use
- Route
- subcutaneous, abdomen or thigh
- Easy mix
- 10 mg + 2 mL = 5 mg/mL, so 1 mg = 0.2 mL = 20 units, 1.75 mg (the label dose) = 35 units and 0.5 mg = 10 units.

About the PT-141 math
PT-141, also listed as Bremelanotide, is a synthetic cyclic seven-amino-acid peptide. Research listings are almost always 10 mg vials, and a few suppliers carry a spray format that this page does not cover. With a single vial size, the only decision is the solvent volume, and 1 mL is the one that gives the roundest numbers for milligram-scale draws. A 10 mg vial with 1 mL of bacteriostatic water is 10 mg/mL, or 10,000 mcg/mL. A 1 mg draw is 0.1 mL, or 10 units on a U-100 syringe, and the vial holds ten draws. With 2 mL the concentration is 5 mg/mL and the same 1 mg draw is 0.2 mL, or 20 units. With 2.5 mL it is 4 mg/mL and 1 mg is 0.25 mL, or 25 units. Smaller draws follow the same ratio: 500 mcg at 10 mg/mL is 5 units, and at 5 mg/mL it is 10 units. The number of draws in the vial, 10,000 mcg divided by the draw size, never depends on the volume.
PT-141 questions
PT-141 dosage for men?
There is no approved male dose. The 2004 dose study found significant erections above 1 mg and used 4 and 6 mg in men who did not respond to sildenafil. Most men start at 0.5 to 1 mg and settle at 1 to 2 mg, because nausea rises fast above that. The female label dose, 1.75 mg, is a sensible ceiling to start under.
How many units of PT-141 is 1 mg?
With 10 mg in 2 mL (5 mg/mL), 1 mg is 20 units on a U-100 syringe. 1.75 mg is 35 units, 2 mg is 40 units. If you used 1 mL of water instead, halve those numbers.
How many units is 1 mg of PT-141 at 10 mg in 1 mL?
10 mg divided by 1 mL is 10 mg/mL. 1 mg divided by 10 mg/mL is 0.1 mL, which is 10 units on a U-100 syringe. The vial holds 10 such draws.
How much bacteriostatic water for a 10 mg vial of PT-141?
Any volume the vial holds. 1 mL gives 10 mg/mL, 2 mL gives 5 mg/mL and 3 mL gives 3.33 mg/mL (rounded). A 500 mcg draw is 5 units, 10 units or 15 units at those three concentrations.
How many 2 mg draws does a 10 mg vial of PT-141 hold?
10 divided by 2 is 5 draws. With 1 mL of solvent (10 mg/mL) each 2 mg draw is 0.2 mL, or 20 units; with 2 mL (5 mg/mL) it is 0.4 mL, or 40 units.
