Why concentration is the number to keep

Once the solvent is in, the concentration is the only number you need to carry forward. Every later question, how many units for a given amount, how many draws the vial holds, what a unit carries, is a one-line division from it. Write it on the vial in mg/mL and in mcg per unit, and the syringe math becomes a lookup.

Concentration also makes vials comparable. A 5 mg vial in 1 mL and a 10 mg vial in 2 mL are both 5 mg/mL and behave identically at the syringe; the only difference is that the second holds twice as many draws. The vial tables list every pairing side by side.

Questions

How do I calculate peptide concentration after reconstitution?

Divide the mass in the vial by the volume of solvent you added. 10 mg in 2 mL is 5 mg/mL. Multiply by 1,000 to express it in mcg/mL: 5,000 mcg/mL.

What is mcg per unit?

The concentration divided by the number of units in a millilitre. At 5,000 mcg/mL on a U-100 syringe, one unit (0.01 mL) holds 50 mcg. On a U-40 syringe one unit (0.025 mL) holds 125 mcg.

Does the powder add volume?

A few milligrams of lyophilized peptide adds a negligible volume to a millilitre or more of solvent, so the arithmetic treats the final volume as the solvent volume. For very large masses in very small volumes the true concentration is slightly lower than the simple division.
